Art-wise, two must-visit spots in Barcelona are MACBA http://www.macba.es/english/index.html, the Museum of Contemporary Art, designed by Richard Meier (who also did the new Getty Museum), and the CCCB http://www.cccb.es/ingles/cccb.htm, the Contemporary Culture Centre, which used to be a hospice in the 18th century. There's also a really good contemporary art gallery next to Gaudi's La Perdrera but its name escapes me for the time being.

Of course, the Guggenheim in Bilbao goes without saying ;-)

Also, look out for b-guided http://b-guided.com/...you can pick up this shopping/art/culture bible to Barcelona when you get there. FABULOUS!

And food? You can't go wrong with a breakfast of spicy sausages, tortilla and coffee at Mercat Boqueria, the massive food market just off La Rambla.

In terms of architecture most people think of Gaudi in BCN but you must visit Mies van der Rohe's Barcelona Pavillon at Placa de Espana. Go there and pay homage.
